Basics and Applications of Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry (MALDI-MS)

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) has become an essential tool in many scientific investigations. In this presentation, the basics of MALDI-MS will be covered and then several applications of MALDI-MS will be presented. In the analysis of small analytes using matrix-assisted laser desorption/ionization (MALDI), unknown compounds were constantly observed on stainless steel plates around m/z 304.3, 332.3, 360.4, and 388.4, regardless of the matrix and/or solvent and even with bare plates. These unknown compounds were successfully characterized to be benzylalkylmethylammonium salts using three different types of MALDI-MS instrumentation: MALDI-TOF MS, MALDI-TOF/TOF MS, and MALDI-FTMS. Recently, the addition of phosphoric acid to the matrix system in MALDI-MS was reported to improve MALDI-MS analysis. The details of the experimental process with phosphoric acid additive will be provided. Finally, MALDI-MS sample preparation using free-vacuum drying where the matrix-analyte mixture will be introduced where the mixture is first frozen and then dried under vacuum, resulted in a homogeneously crystallized matrix-analyte mixture appropriate for quantitative MALDI-MS analysis, improving the reproducibility of MALDI-MS analysis.
