Chemical biology of modified oligonucleotides: SNP detection and gene regulation

Sensing single mutation and manipulating target gene expression are two important areas in nucleic acid chemical biology. We developed a group of fluorescent oligonucleotide probes by site-specific incorporation of pyrene modified thymidine phosphoramidite. These new types of pyrene labeled fluorescence oligonucleotide probes were very sensitive and selective for single nucleotide polymorphism (SNP) and were successfully used for the detection of BRAF single mutation site of human melanoma. In addition, we also developed a group of photolabile oligonucleotides for spatiotemporal regulation of target gene expression with circular antisense oligonucleotides or siRNAs site-specifically labelled with caged functional groups.
